Overview

The combination of cyclophosphamide, lenalidomide, and dexamethasone is a multi-agent chemotherapy regimen primarily used in the treatment of multiple myeloma. Cyclophosphamide is an alkylating agent that interferes with DNA replication and cell division. Lenalidomide is an immunomodulatory drug that exerts anti-myeloma effects through direct cytotoxicity, inhibition of angiogenesis, and modulation of the immune system. Dexamethasone is a corticosteroid that enhances the efficacy of chemotherapy by reducing inflammation and directly inducing apoptosis in myeloma cells. This triplet therapy has demonstrated high response rates and good tolerability both as initial therapy for newly diagnosed multiple myeloma patients and in relapsed/refractory settings[1][2][3][4]. The combination leverages synergistic mechanisms to improve outcomes compared to doublet regimens.
