This is a multi-agent chemotherapy combination consisting of five drugs: cyclophosphamide (an alkylating agent), methotrexate (an antimetabolite and folic acid antagonist), fluorouracil (a pyrimidine analog antimetabolite), vincristine (a vinca alkaloid that inhibits microtubule formation), and prednisone (a synthetic glucocorticoid corticosteroid). The regimen is used primarily for the treatment of various solid tumors including breast cancer, lung cancer, ovarian cancer, and gastrointestinal cancers. Each component acts through a distinct mechanism to inhibit tumor cell proliferation or induce apoptosis. Cyclophosphamide crosslinks DNA; methotrexate inhibits dihydrofolate reductase blocking nucleotide synthesis; fluorouracil interferes with DNA/RNA synthesis by inhibiting thymidylate synthase; vincristine disrupts mitotic spindle formation by binding tubulin; prednisone induces apoptosis in certain lymphoid cells. This combination has been shown to improve response rates in several solid tumors but is associated with moderate to severe toxicity in a significant proportion of patients[1][4].
