Overview

This is a combination therapy consisting of **cyclophosphamide**, an alkylating agent used for immunomodulation, and **SB-728-T**, an investigational autologous cell-based gene therapy. SB-728-T involves ex vivo modification of a patient's autologous CD4+ T cells by disrupting the gene encoding the CCR5 receptor using zinc finger nuclease (ZFN) genome-editing technology, rendering the cells resistant to HIV infection. Cyclophosphamide is used as a lymphodepleting pre-conditioning agent to enhance the engraftment, persistence, and proliferation of the modified T cells when reinfused into the patient. This therapeutic strategy is being developed and studied primarily for HIV/AIDS as a potential "functional cure" by achieving long-term viral control without antiretroviral therapy[1][2][3][4].
