Cyclophosphamide + somatostatin is an experimental combination therapy consisting of the alkylating agent cyclophosphamide and the peptide hormone somatostatin. Cyclophosphamide is a small molecule antineoplastic and immunosuppressive agent that acts as a DNA-alkylating agent, leading to cell death, particularly in rapidly dividing cells. Somatostatin is a peptide hormone that binds to somatostatin receptors (SSTRs), inhibiting adenylyl cyclase activity and reducing secretion of various hormones and growth factors. The combination has been investigated in phase II clinical trials for low-grade non-Hodgkin's lymphoma, chronic lymphocytic leukemia, advanced breast cancer, and advanced lung adenocarcinoma—often as part of multidrug regimens with other agents such as bromocriptine, retinoids, melatonin, ACTH or vitamin D3[1][3][4][6]. The rationale for this combination includes both direct cytotoxic effects (cyclophosphamide) and modulation of tumor growth via hormonal pathways (somatostatin). Reported studies suggest mild toxicity profiles with some evidence of partial or complete responses in heavily pretreated or advanced-stage patients.
