Cyclophosphamide + viagenpumatucel-l is a combination therapy investigated for the treatment of advanced non-small cell lung cancer (NSCLC). Cyclophosphamide is a well-established alkylating agent used in chemotherapy, while viagenpumatucel-l (HS-110) is an investigational allogeneic cellular cancer vaccine. Viagenpumatucel-l consists of a human lung adenocarcinoma cell line engineered to express multiple tumor antigens (such as MAGE-A3, NY-ESO-1, LAGE-1) and secrete a modified heat shock protein gp96-Ig. This protein chaperones tumor antigens and delivers them directly to antigen-presenting cells via the cross-presentation pathway, leading to selective activation of CD8+ cytotoxic T cells. The combination aims to enhance anti-tumor immune responses by both direct cytotoxic effects from cyclophosphamide and immune priming from the vaccine[1][3][5][6]. The primary indication studied was advanced NSCLC in patients who had failed at least two prior lines of therapy.
