Cycloserine + tacrolimus is a combination of two pharmaceutical agents with distinct mechanisms and clinical uses. Cycloserine is an antibiotic primarily used in the treatment of multidrug-resistant tuberculosis, acting as a structural analog of D-alanine to inhibit cell wall synthesis in bacteria. Tacrolimus is an immunosuppressant, classified as a calcineurin inhibitor, widely used to prevent organ transplant rejection and treat certain autoimmune conditions. Tacrolimus binds to FK506 binding protein 12 (FKBP-12), forming a complex that inhibits calcineurin phosphatase activity, thereby blocking T-cell activation by preventing the transcription of interleukin-2 and other cytokines[1][3][4][5][6]. The combination does not represent a standard or approved therapy; there are no known indications for their combined use in clinical practice.
