This is a combination regimen consisting of four drugs: - **Cyclosporine** is an immunosuppressant that inhibits calcineurin, thereby suppressing T-cell activation and cytokine production. - **Cytarabine** is a nucleoside analog antimetabolite that inhibits DNA synthesis, primarily used in the treatment of acute myeloid leukemia (AML) and other hematologic malignancies. - **Daunorubicin** is an anthracycline antibiotic with antimitotic and cytotoxic activity. It intercalates into DNA, inhibits topoisomerase II, generates free radicals, and disrupts nucleic acid synthesis[1][3][5][7]. - **Gemtuzumab ozogamicin** is an antibody-drug conjugate composed of a humanized anti-CD33 monoclonal antibody linked to calicheamicin (a cytotoxic agent). It targets CD33-positive leukemic cells in AML; after binding to CD33 on the cell surface, it delivers calicheamicin intracellularly to induce double-stranded DNA breaks and apoptosis[6][8][10]. This combination may be used as part of investigational or intensive induction regimens for certain subtypes of acute myeloid leukemia (AML), particularly those expressing CD33.
