CD33 is a transmembrane receptor primarily expressed on cells of the myeloid lineage. It functions as an inhibitory receptor that modulates immune cell activity by binding to sialic acid residues. Upon ligand binding, ITIM motifs are phosphorylated, leading to the recruitment of SHP phosphatases and suppression of cellular activation functions such as phagocytosis. CD33 is a therapeutic target in Acute Myeloid Leukemia, and genetic polymorphisms affecting its expression have been linked to Alzheimer's disease risk.
Upon ligand binding, ITIM motifs are phosphorylated, recruiting SHP phosphatases which initiate an inhibitory signal cascade that suppresses cellular activation functions.
