Cyclosporine + methylprednisolone is a combination of two immunosuppressive drugs used primarily for the treatment of autoimmune diseases and in organ transplantation. Cyclosporine is a calcineurin inhibitor that suppresses T-cell mediated immune responses by inhibiting the activity of calcineurin, thereby reducing interleukin-2 production and T-cell proliferation. Methylprednisolone is a synthetic glucocorticoid corticosteroid that exerts anti-inflammatory and immunosuppressive effects by modulating gene expression to decrease the production of pro-inflammatory cytokines and mediators. This combination has been studied as second-line therapy for conditions such as severe alopecia areata, systemic lupus erythematosus (SLE), and for prevention or treatment of graft-versus-host disease (GVHD) after transplantation[1][2][4]. The rationale behind combining these agents is to achieve synergistic immunosuppression while potentially allowing lower doses of each drug, which may reduce side effects compared to higher-dose monotherapy.
