Drug intelligence / Profile preview
Cyclosporine (specifically the formulation developed by Zhaoke Ophthalmology) is a calcineurin inhibitor immunosuppressant used for the treatment of moderate to severe dry eye disease. It works by binding to cyclophilin within T-lymphocytes, forming a complex that inhibits the phosphatase activity of calcineurin. This inhibition prevents the translocation of the nuclear factor of activated T-cells (NFAT), thereby suppressing the production of pro-inflammatory cytokines such as interleukin-2 (IL-2). In the context of dry eye disease, this mechanism reduces immune-mediated inflammation of the ocular surface and lacrimal glands, promoting natural tear production. Zhaoke's formulation is a 0.05% ophthalmic gel designed to improve the bioavailability and residence time of the drug on the eye compared to traditional emulsions, potentially allowing for improved efficacy and patient comfort.
