Prednisporin (FOV1101) is a topical ophthalmic fixed-dose combination therapy consisting of low-dose prednisolone acetate and cyclosporine A. Developed by Fovea Pharmaceuticals (later acquired by Sanofi), it is designed to treat persistent allergic conjunctivitis and chronic inflammatory conjunctivitis. Prednisolone acetate is a corticosteroid that acts as a glucocorticoid receptor agonist to suppress inflammation, while cyclosporine A is an immunosuppressant that inhibits calcineurin-mediated T-cell activation. The combination is formulated to maintain the anti-inflammatory efficacy of potent steroids while providing an improved safety profile, specifically aiming to avoid the increase in intra-ocular pressure typically associated with high-dose corticosteroid use. The drug was originally discovered by CombinatoRx using its proprietary combination discovery platform and licensed to Fovea for ophthalmic indications.
