CyGenica's novel drug conjugate is an investigational therapeutic candidate being developed for the treatment of glioblastoma multiforme (GBM). The conjugate utilizes CyGenica's proprietary GEENIE (enablinG safE and targEted iNtracellular delIvEry) platform, which features a 48-KDa engineered, negatively-charged protein that functions as a "molecular drill" or "nanoneedle." This delivery system employs a unique non-endocytic helical mechanism to penetrate cell membranes and deliver therapeutic cargo—including small molecules, nucleic acids, and gene-editing components—directly into the cytosol or nucleus without causing cellular damage. By bypassing the endosomal pathway, the platform aims to overcome endosomal trapping, thereby enhancing the efficacy and safety of the delivered therapeutics. The drug candidate received FDA Orphan Drug Designation for the treatment of GBM in September 2023 and is currently in pre-clinical development.
