Overview

CYNK-001 + aldesleukin is an investigational combination therapy comprised of CYNK-001, a cryopreserved, allogeneic, off-the-shelf natural killer (NK) cell therapy candidate derived from placental CD34+ hematopoietic stem cells, and aldesleukin, a recombinant form of interleukin-2 (IL-2). CYNK-001 is designed to harness the innate anti-tumor activity of NK cells, which express activating receptors (NKG2D, CD94, DNAM1, NKp30, NKp44, NKp46) and cytotoxic effectors such as perforin and granzyme B. Aldesleukin is administered as a cytokine support to stimulate NK cell proliferation, survival, and function. The combination is being developed primarily for treatment-resistant hematologic malignancies, particularly relapsed/refractory acute myeloid leukemia (R/R AML) and measurable residual disease-positive (MRD+) AML. Celularity is developing CYNK-001 and conducting early-phase clinical trials; aldesleukin is a marketed product (recombinant human interleukin-2) used in other settings, repurposed here to augment NK cell therapy efficacy[1][2].
