CyPep-1 is a chemically synthesized peptide of 27 amino acids developed as a first-in-class bifunctional immunotherapy for the treatment of solid tumors. It exhibits a unique dual mechanism of action by both directly lysing cancer cell membranes (oncolytic activity) and inhibiting the oncogenic Wnt/β-catenin signaling pathway through activation of Axin2. This results in tumor-specific cell destruction and immune activation via neoantigen release into the tumor microenvironment. The drug is being investigated primarily for cancers characterized by aberrant Wnt/β-catenin signaling—including adrenocortical carcinoma (ACC), triple-negative breast cancer (TNBC), malignant melanoma, squamous cell carcinoma, and other solid tumors. Preclinical studies have shown that CyPep-1 can synergize with anti-PD-1 therapies such as pembrolizumab to enhance anti-tumor responses[2][5][7][6].
