CYR212 is an engineered IgG-degrading enzyme (IgG protease) developed by Cyrus Biotechnology for the treatment of chronic IgG-driven autoimmune diseases, including myasthenia gravis and immune thrombocytopenia. It is a next-generation version of the bacterial IdeS (imlifidase) enzyme, designed using AI to have an extended half-life and reduced immunogenicity compared to wild-type IdeS. CYR212 rapidly depletes pathogenic IgG autoantibodies, providing rapid and sustained relief from symptoms in diseases where IgG antibodies drive pathology. Unlike earlier versions such as imlifidase, CYR212 demonstrates improved stability, manufacturability, predictable reduction of IgG levels in preclinical models, and does not induce anti-drug antibodies after repeated dosing. Its mechanism involves enzymatic cleavage of human immunoglobulin G (IgG), thereby reducing circulating pathogenic antibodies implicated in various autoimmune conditions[1][2][6].
