A Cyr61 neutralizing antibody is a biologic agent, typically a monoclonal antibody, designed to bind to and inhibit the activity of Cysteine-Rich Angiogenic Inducer 61 (CYR61), also known as CCN1. CYR61 is a secreted heparin-binding protein that plays a critical role in cell proliferation, migration, and angiogenesis. In oncology, CYR61 is often overexpressed and associated with tumor progression, metastasis, and resistance to chemotherapy in various cancers, including breast cancer, pancreatic cancer, and acute myeloid leukemia (AML). By neutralizing CYR61, these antibodies inhibit downstream signaling pathways such as AKT and ERK, potentially normalizing tumor vasculature and suppressing metastatic growth. Currently, these antibodies are primarily used as research tools and have not yet entered formal clinical development as therapeutic agents.
