Overview

CYT-0851 + gemcitabine is an investigational combination therapy under clinical evaluation for advanced solid tumors and certain hematologic malignancies. CYT-0851 is a small molecule inhibitor of monocarboxylate transporters (MCTs), which blocks lactate transport, leading to glycolytic cancer cell death. It also acts as a homologous recombination inhibitor by targeting RAD51-mediated DNA repair, inducing synthetic lethality in cancer cells[2][3][6]. Gemcitabine is an antimetabolite chemotherapy agent that inhibits DNA synthesis and is approved for various cancers including pancreatic, lung, breast, and ovarian cancers[1]. The combination has shown early clinical activity and a generally well-tolerated safety profile in heavily pretreated patients with advanced solid tumors[3][5].
