CYT-150 is an investigational, induced pluripotent stem cell (iPSC)-derived natural killer (iNK) cell therapy being developed by Cytovia Therapeutics for the treatment of multiple myeloma. The therapy is engineered using TALEN® gene-editing technology, through a collaboration with Cellectis, to eliminate the expression of CD38 on the surface of the NK cells. This CD38 knockout is a critical modification designed to prevent 'fratricide'—the self-destruction of NK cells that occurs when they are administered alongside CD38-targeting monoclonal antibodies like daratumumab or isatuximab. By preventing fratricide, CYT-150 aims to maximize the synergistic anti-tumor effects of both the exogenous NK cells and the antibody-dependent cellular cytotoxicity (ADCC) promoted by the co-administered treatment, thereby improving persistence and efficacy in patients with relapsed or refractory multiple myeloma.
