Overview

CYT-303 is a multifunctional bispecific antibody designed as a natural killer (NK) cell engager for cancer immunotherapy. Developed using Cytovia’s proprietary FLEX-NK platform, it simultaneously targets the NK cell activating receptor NKp46 and the tumor antigen Glypican-3 (GPC3), which is highly expressed in hepatocellular carcinoma (HCC) and other solid tumors but minimally present in normal adult tissues. The molecule incorporates humanized binders for both GPC3 and NKp46, enabling potent redirection of NK cells to kill GPC3-expressing tumor cells. Preclinical studies demonstrate that CYT-303 mediates strong cytotoxicity against HCC tumor lines, reverses dysfunction in serially activated NK cells, and induces antibody-dependent cellular phagocytosis (ADCP) as well as complement-dependent cytotoxicity (CDC). In animal models, it showed no toxicity or cytokine release at high doses administered intravenously. The drug is currently in preclinical development for hepatocellular carcinoma and other solid tumors[1][2][3][8].
