CYT X400 is a preclinical-stage, multispecific pan-gamma delta (γδ) T-cell engager (TCE) being developed by Cytospire Therapeutics for the treatment of solid tumors. The therapeutic is designed to simultaneously bind an undisclosed tumor-associated antigen (TAA) on tumor cells and a shared epitope on the gamma-delta T-cell receptor (γδ TCR) across all γδ T-cell subsets, including tissue-resident Vδ1 and Vδ3 cells and blood-resident Vδ2 cells. By redirecting and activating these unconventional T cells to the tumor site, CYT X400 promotes potent, MHC-independent tumor cell lysis. This pan-γδ engagement strategy is intended to overcome patient heterogeneity and offer an improved safety profile with a lower risk of cytokine release syndrome (CRS) compared to conventional CD3-targeting T-cell engagers.
