Overview

CYT003-QbG10 is an investigational immunotherapeutic drug developed for the treatment of allergic diseases such as allergic rhinoconjunctivitis and asthma. It is based on Cytos Biotechnology's Immunodrug platform and consists of a virus-like particle (Qb) loaded with a synthetic immunostimulatory DNA sequence (G10), which acts as a toll-like receptor 9 (TLR9) agonist. The mechanism aims to modulate the immune response by shifting it away from Th2-dominated pathways associated with allergy, towards Th1 responses, thereby reducing IgE-mediated symptoms without using allergens themselves. This allergen-independent approach seeks to improve safety and tolerability compared to traditional allergen-based immunotherapies[1][3][5][6]. Clinical studies have shown that high-dose CYT003-QbG10 can reduce symptoms and medication use in allergic rhinoconjunctivitis[1], maintain asthma control during corticosteroid withdrawal in some patients[6][8], but failed to show efficacy over placebo in larger phase II trials for moderate-to-severe allergic asthma, leading to termination of further development for this indication[4][5].
