**CYT004-MelQbG10** is an investigational therapeutic melanoma vaccine developed by Cytos Biotechnology for HLA-A*0201-positive patients with stage II to IV malignant melanoma. It is a bacteriophage Qbeta-derived virus-like nanoparticle containing the A-type CpG oligodeoxynucleotide G10 and covalently coupled to a Melan-A/MART-1 peptide spanning amino acids 16 to 35. The vaccine is designed to promote dendritic-cell uptake and cross-presentation of the Melan-A antigen while CpG G10 activates Toll-like receptor 9, inducing antigen-specific cytotoxic CD8-positive T-cell responses against Melan-A-expressing melanoma cells. Phase I/II and phase IIa studies reported acceptable tolerability and vaccine-induced Melan-A-specific T-cell responses; Montanide ISA-51 and topical imiquimod were evaluated as separately administered adjuvant approaches in some trial arms.
