Cytarabine + glasdegib is a fixed combination regimen used primarily for the treatment of newly diagnosed acute myeloid leukemia (AML) in adults who are 75 years or older or who have comorbidities that preclude intensive induction chemotherapy. Cytarabine is a nucleoside analog antimetabolite that inhibits DNA synthesis, while glasdegib is a selective small-molecule inhibitor of the Hedgehog signaling pathway, acting specifically by binding to and inhibiting Smoothened (SMO), a transmembrane protein involved in Hedgehog signal transduction. The combination has demonstrated improved overall survival compared to low-dose cytarabine alone in this patient population[1][2][6]. The regimen is administered as oral glasdegib with subcutaneous low-dose cytarabine.
