Smoothened (SMO) is a transmembrane protein and a member of the Class Frizzled (Class F) G protein-coupled receptor (GPCR) family. It plays an essential role in the Hedgehog (Hh) signaling pathway, which is crucial for embryonic development, tissue patterning, and adult tissue homeostasis. Dysregulation of Hedgehog/SMO signaling can result in developmental disorders or contribute to oncogenesis, such as basal cell carcinoma and medulloblastoma.
SMO antagonists inhibit SMO, preventing activation of the Hedgehog pathway.
