The core backbone of this cycle consists of high-dose cytarabine (HD-ARAC) and pegylated asparaginase (pegaspargase). Cytarabine is an antimetabolite that inhibits DNA synthesis by competing with cytidine for incorporation into DNA and inhibiting DNA polymerase. Pegaspargase is an enzyme that depletes systemic L-asparagine, an amino acid essential for the survival of leukemic blasts that lack asparagine synthetase.
