Overview

This combination therapy consists of the pyrimidine analog **cytarabine** and the selective Bruton's tyrosine kinase (BTK) inhibitor **zanubrutinib**. Cytarabine acts as an antimetabolite that inhibits DNA polymerase and incorporates into DNA, leading to cell death during the S-phase of the cell cycle. Zanubrutinib is a small-molecule inhibitor that covalently binds to BTK, blocking the B-cell receptor signaling pathway which is often constitutively active in B-cell malignancies. This combination is primarily being investigated by researchers at Huashan Hospital, Fudan University, for the treatment of primary central nervous system lymphoma (PCNSL), aiming to leverage the CNS-penetrating properties of both agents to improve outcomes in this aggressive disease.
