Cytokine-activated natural killer (NK) cells are an adoptive cellular immunotherapy involving the ex vivo expansion and activation of NK cells using specific cytokines, such as Interleukin-2 (IL-2), Interleukin-12 (IL-12), Interleukin-15 (IL-15), or Interleukin-18 (IL-18). This process enhances the cells' proliferative capacity, survival, and cytotoxic effector functions against malignant or virally infected cells. Unlike CAR-NK cells, these are typically non-genetically modified and rely on their natural repertoire of activating receptors (e.g., NKG2D, NKp30, NKp44, NKp46) and the release of cytotoxic granules containing perforin and granzymes to induce target cell apoptosis. They are investigated for the treatment of various solid tumors, hematologic cancers, and chronic viral infections like Hepatitis C and Cytomegalovirus.
