This investigational adoptive cellular immunotherapy, developed by A.O. Ospedale Papa Giovanni XXIII, utilizes donor-derived cytokine-induced killer (CIK) cells for the treatment of hematologic malignancies relapsing after allogeneic stem cell transplantation. CIK cells are ex vivo expanded and activated T lymphocytes produced by the sequential incubation of peripheral blood mononuclear cells with interferon-gamma (IFN-γ), anti-CD3 monoclonal antibody (OKT3), and interleukin-2 (IL-2). The resulting cell population is primarily composed of CD3+/CD56+ natural killer T cells, which possess both T cell and natural killer (NK) cell properties, enabling MHC-unrestricted cytotoxicity against tumor cells. The therapy is typically evaluated in a sequential regimen following unmanipulated donor lymphocyte infusions (DLI) to enhance the graft-versus-leukemia effect while minimizing the risk of graft-versus-host disease.
