relatilmab-Washington University School of Medicine-melanoma refers to a specific investigational combination immunotherapy regimen being evaluated in a Phase 1 clinical trial (NCT05327920) sponsored by the Washington University School of Medicine. This regimen combines cytokine-induced memory-like natural killer (ML NK) cells with two immune checkpoint inhibitors: nivolumab (a PD-1 antagonist) and relatlimab (a LAG-3 antagonist). The ML NK cells are human natural killer cells that have been pre-activated ex vivo with a cocktail of cytokines (IL-12, IL-15, and IL-18) to induce a stable memory-like phenotype characterized by enhanced effector functions and persistence. The combination aims to overcome resistance to standard checkpoint inhibitors in patients with advanced or metastatic melanoma by leveraging the synergistic anti-tumor activity of pre-activated NK cells and dual checkpoint blockade.
