Cytosporone B (Csn-B) is a naturally occurring small molecule polyketide and the first known agonist of the nuclear receptor NR4A1 (Nur77). It binds with high affinity to the ligand-binding domain of Nur77 (IC50 = 0.278 nM), activating Nur77-dependent gene transcription and promoting its translocation from the nucleus to mitochondria, leading to induction of apoptosis and inhibition of cancer cell growth. Csn-B also reduces neuroinflammation by suppressing Th1 and Th17 cell differentiation and inflammatory cytokine production in experimental autoimmune encephalomyelitis (EAE, a model of multiple sclerosis). Additionally, it has demonstrated antiviral activity in a mouse model of influenza A infection, lowering viral loads and improving survival. Csn-B is being explored as a research tool and potential lead compound for neuroinflammatory, viral, metabolic, and neoplastic diseases[1][3][4][6][7][8][9][10].
