Overview

D-4559 is a preclinical macrophage-switching nanomedicine developed by Ashvattha Therapeutics for the treatment of hepatocellular carcinoma (HCC). It belongs to a class of compounds known as 'Dendranibs,' which consist of tyrosine kinase inhibitors (TKIs) conjugated to a proprietary hydroxyl dendrimer (HD) platform. D-4559 specifically utilizes a sorafenib backbone to target and inhibit vascular endothelial growth factor receptor (VEGFR) tyrosine kinases in tumor-associated macrophages (TAMs). This targeted approach is designed to reprogram TAMs from an immunosuppressive M2 phenotype to a pro-inflammatory M1 phenotype, thereby enhancing the immune system's ability to eliminate cancer cells and reducing tumor growth.
