Overview

d-amphetamine + atomoxetine is a combination of two pharmacologically distinct agents used primarily in the management of attention deficit hyperactivity disorder (ADHD). d-Amphetamine is a central nervous system stimulant that increases synaptic concentrations of dopamine and norepinephrine by promoting their release and inhibiting reuptake. Atomoxetine is a selective norepinephrine reuptake inhibitor (NRI) that increases norepinephrine and, to some extent, dopamine levels in the prefrontal cortex by blocking the presynaptic norepinephrine transporter. This combination may be considered in clinical practice for patients with suboptimal response to monotherapy or during transitions between medications. The combination is not formally approved by regulatory agencies but has been studied for its potential to improve ADHD symptoms when monotherapy is insufficient[1][3][7]. Atomoxetine may also attenuate some physiological effects of amphetamines, such as blood pressure elevation[2].
