Drug intelligence / Profile preview
D-PLEX 100 is a locally administered, prolonged-release formulation of the antibiotic doxycycline hyclate, developed by PolyPid Ltd. using its proprietary PLEX (Polymer-Lipid Encapsulation matriX) technology. The drug is designed to be applied directly into the surgical site during surgery, where it provides a high, localized concentration of the antibiotic for up to 30 days to prevent surgical site infections (SSIs). This localized delivery approach aims to maintain effective therapeutic levels at the site of potential infection while keeping systemic exposure low, thereby reducing the risk of systemic side effects and the potential for antibiotic resistance. D-PLEX 100 is currently in Phase 3 clinical development, specifically the SHIELD II trial, focusing on the prevention of infections following elective colorectal surgery and cardiac surgery.
