Overview

d-threo-1-phenyl-2-decanoylamino-3-morpholino-1-propanol (PDMP) is a small molecule drug that acts as an inhibitor of glucosylceramide synthase (GCS). By inhibiting GCS, PDMP can sensitize cancer cells to chemotherapeutic agents like sorafenib and doxorubicin. It has been shown to revert sorafenib resistance in hepatocellular carcinoma by targeting GCS upregulation and to eliminate the oncogenic function of p53 R273H mutant in colon cancer cells, restoring wild-type p53 expression and decreasing cancer stem cells. Its mechanism involves triggering mitochondrial cell death after energetic collapse and impacting epithelial-mesenchymal transition and induced pluripotency of cancer stem cells.
