Overview

D-TTM020 is a dendrimer-based glutaminase (GLS1) inhibitor designed to selectively target microglial GLS1. Developed by researchers at Johns Hopkins University, it is being investigated for the treatment of major depressive disorder (MDD). By inhibiting the upregulation of GLS1 in activated microglia, D-TTM020 aims to mitigate the neuroinflammatory pathophysiology associated with chronic stress and depression. In preclinical mouse models of chronic social defeat stress (CSDS), D-TTM020 demonstrated the ability to reduce microglial GLS1 activity in the hippocampus and prefrontal cortex, leading to significant improvements in sociability, cognition, and exploratory behavior. The dendrimer delivery system is intended to enhance microglial targeting while maintaining a favorable gastrointestinal safety profile compared to non-targeted inhibitors.
