Overview

D0405 scFv CAR-T is an experimental second-generation chimeric antigen receptor (CAR) T-cell therapy targeting mesothelin (MSLN), a cell-surface glycoprotein overexpressed in various solid malignancies including pancreatic, ovarian, and lung cancers. Developed by Lentigen Technology (a Miltenyi Biotec company), the construct features a single-chain variable fragment (scFv) specific to the membrane-proximal region of MSLN, paired with a CD28 co-stimulatory domain and a CD3ζ signaling domain. This design aims to maintain potency in the presence of shed MSLN, which often hinders CAR-T efficacy in clinical settings. Preclinical data presented at AACR 2025 demonstrated that D0405 exhibits high surface expression, robust in vitro cytotoxicity, and significant cytokine production (TNFα, IL-2, and IFNγ) upon target engagement.
