D07001-F4 is an oral formulation of the pyrimidine nucleoside analog gemcitabine, developed by InnoPharmax using its proprietary OralPas platform. The OralPas technology is a self-microemulsifying drug delivery system (SMEDDS) designed to enhance the oral bioavailability of drugs that typically have poor absorption or high gastrointestinal degradation. Gemcitabine is a standard-of-care antimetabolite usually administered intravenously; D07001-F4 aims to provide a more convenient oral alternative with potentially improved safety and efficacy profiles through modified pharmacokinetics. Once absorbed, gemcitabine is phosphorylated into active metabolites that inhibit DNA synthesis by blocking ribonucleotide reductase and by competing with cytidine for incorporation into DNA strands, leading to chain termination and apoptosis. It is currently being investigated in clinical trials for the treatment of advanced solid tumors, including cholangiocarcinoma, and malignant lymphomas.
