D3L-001 is a novel bispecific antibody that targets both the human epidermal growth factor receptor 2 (HER2, also known as ErbB2) and the cell surface antigen CD47. By simultaneously binding to HER2 on tumor cells and CD47, which acts as a "don't eat me" signal to immune cells, D3L-001 is designed to stimulate immune-mediated phagocytosis and cytotoxicity against cancer cells. This dual targeting approach aims to enhance antitumor activity by promoting both direct tumor cell killing and immune system activation. The drug is being developed primarily for HER2-positive cancers, including breast cancer, gastric cancer, gastroesophageal junction carcinomas, and other solid tumors. Preclinical studies have shown potent synergistic antitumor effects via HER2-guided CD47 co-blockade in vitro and in vivo models[1][4][5][8].
