D561-0775 is a novel, direct small molecule activator of 5'-adenosine monophosphate-activated protein kinase (AMPK), specifically targeting the AMPK α subunit. Identified through in silico virtual screening by researchers at the Macau University of Science and Technology, it has demonstrated significant inhibitory effects against gefitinib-resistant non-small cell lung cancer (NSCLC) cell lines, such as H1975. The compound works by activating the AMPK/mTOR signaling pathway, which leads to the induction of apoptosis, cell cycle arrest, and the suppression of glycolysis and cholesterol synthesis. Its ability to inhibit lipid metabolism makes it a potential therapeutic candidate for overcoming EGFR-TKI resistance in lung cancer patients.
