Overview

D7 is a preclinical-stage small molecule molecular glue degrader developed by Isoprene Pharmaceuticals. It is a dual-action agent designed to induce the proteasomal degradation of both Mitogen-Activated Protein Kinase Interacting Kinases 1 and 2 (MNK1/2) and the Androgen Receptor (AR). By degrading MNK1/2, D7 prevents the phosphorylation of eukaryotic translation initiation factor 4E (eIF4E), thereby inhibiting the translation of oncogenic mRNAs. Concurrently, the degradation of AR (including variants such as AR-V7) targets the primary driver of prostate cancer growth and addresses resistance mechanisms in hormone-dependent cancers. This bifunctional degradative approach is primarily intended to overcome resistance in advanced oncology indications, specifically prostate cancer and breast cancer.
