DA + CPT-KK-3E8 is an experimental, targeted nanoparticle delivery system designed for the treatment of cancers expressing Tumor-Associated Glycoprotein 72 (TAG-72). The platform consists of a self-assembled nanotube core formed by camptothecin conjugated to a dilysine peptide (CPT-KK), which is stabilized by a layer of polydopamine (PDA). The surface of this PDA-stabilized nanotube is functionalized with 3E8, a single-chain variable fragment (scFv) that specifically targets the TAG-72 antigen, which is overexpressed in various adenocarcinomas, including colorectal, ovarian, and breast cancers. Once the nanoparticle binds to and is internalized by the target cancer cell, the camptothecin payload acts as a potent DNA topoisomerase I inhibitor, inducing double-strand DNA breaks and subsequent apoptosis. This modular platform is designed to improve the solubility and therapeutic index of camptothecin while minimizing systemic toxicity through active tumor targeting.
