Overview

KNP-101 is a bi-specific immunocytokine designed for cancer immunotherapy, particularly targeting solid tumors. It consists of an engineered interleukin-12 (IL-12) mutant arm with attenuated activity to reduce toxicity, fused to an antibody that targets fibroblast activation protein (FAP), which is highly expressed in the tumor microenvironment (TME) of many solid tumors. This dual approach aims to deliver IL-12 selectively to the TME, enhancing anti-tumor immune responses while minimizing systemic side effects. The drug is being developed as a next-generation therapy for use alone or in combination with checkpoint inhibitors, especially in non-small cell lung cancer (NSCLC) and other FAP-expressing solid tumors. Its current development stage is preclinical[1][2][7].
