DA-4507 is a preclinical-stage small molecule inhibitor developed by Dong-A ST that targets General Control Nonderepressible 2 (GCN2) kinase, also known as EIF2AK4. GCN2 acts as a metabolic sensor that is activated in response to amino acid deprivation within the tumor microenvironment, a condition frequently caused by the overexpression of enzymes such as IDO1 or arginase. Upon activation, GCN2 phosphorylates the alpha subunit of eukaryotic initiation factor 2 (eIF2α), which triggers the integrated stress response, promoting tumor cell survival under metabolic stress and inducing an immunosuppressive environment that inhibits T-cell proliferation and function. By inhibiting GCN2, DA-4507 aims to block these adaptive survival mechanisms and restore anti-tumor immune activity, potentially serving as a novel therapeutic approach in oncology.
