DA-4515 is a preclinical-stage small molecule inhibitor developed by Dong-A ST that targets the TEA Domain (TEAD) transcription factors. As a key component of the Hippo signaling pathway, TEAD proteins interact with co-activators YAP (Yes-associated protein) and TAZ (transcriptional co-activator with PDZ-binding motif) to regulate genes involved in cell proliferation, survival, and apoptosis. Dysregulation of the Hippo pathway is a common driver in various cancers, including those with NF2 mutations or YAP/TAZ amplification, leading to uncontrolled tumor growth. DA-4515 is designed to disrupt the TEAD-mediated transcriptional complex, thereby providing a potential therapeutic approach for solid tumors that are dependent on Hippo pathway signaling.
