DA7 is an immunotoxin composed of the mouse IgG2b monoclonal antibody 3A1E, which targets the CD7 antigen, chemically linked to deglycosylated ricin A-chain (dgA). CD7 is a cell surface glycoprotein expressed on the majority of T-cell lineage leukemia and lymphoma cells. Once the immunotoxin binds to CD7 and is internalized, the ricin A-chain enzymatically cleaves the N-glycosidic bond of a specific adenine residue in the 28S ribosomal RNA of the 60S subunit. This action irreversibly inhibits protein synthesis, leading to cell death. DA7 was developed for the treatment of refractory T-cell acute lymphoblastic leukemia (T-ALL) and has shown potent efficacy in disseminated human leukemia models in SCID mice.
