Dab2 mRNA nanoparticle is an experimental mRNA-based therapeutic consisting of mRNA encoding the Disabled Homolog 2 (Dab2) protein encapsulated within lipid nanoparticles (LNPs). Dab2 is an endocytic adaptor protein that plays a critical role in regulating the trafficking and signaling of receptor tyrosine kinases, specifically Vascular Endothelial Growth Factor Receptor 2 (VEGFR2). In preclinical models of diabetic wound healing, the delivery of Dab2 mRNA promotes angiogenesis and accelerates tissue repair. In the context of atherosclerosis, Dab2 expression in the endothelium is regulated by the transcription factor KLF4 and is essential for maintaining endothelial homeostasis through the PI3K-Akt-eNOS signaling pathway. Targeted delivery of Dab2 mRNA to the atherogenic endothelium has been shown to restrain plaque progression and promote atheroma regression in mouse models.
