Overview

**DAB486-G-CSF** is an experimental recombinant fusion toxin comprising a 486-amino-acid diphtheria toxin-derived cytotoxic domain genetically linked to human granulocyte colony-stimulating factor. The G-CSF domain targets cells expressing the granulocyte colony-stimulating factor receptor, including G-CSF-responsive acute myeloid leukemia cells, enabling uptake of the toxin moiety. Intracellular diphtheria-toxin enzymatic activity ADP-ribosylates elongation factor 2, thereby inhibiting protein synthesis and causing cell death. In preclinical studies, the construct was cytotoxic to G-CSF-dependent OCI/AML1 cells and inhibited colony formation by G-CSF-responsive leukemic blasts from a patient with acute myeloid leukemia, while sparing factor-independent OCI/AML3 cells.
