Overview

DAB486IL-2 is a recombinant fusion toxin engineered by replacing the receptor-binding domain of diphtheria toxin with human interleukin-2 (IL-2). This design allows the molecule to selectively bind and intoxicate cells expressing high-affinity interleukin-2 receptor (IL-2R) (primarily activated T cells and certain hematologic malignancies). The drug acts by delivering the cytotoxic action of diphtheria toxin specifically to IL-2R-positive cells, leading to their death. It was developed as a targeted therapy for diseases such as refractory rheumatoid arthritis and chemotherapy-resistant hematologic malignancies. Clinical trials demonstrated selective cytotoxicity for IL-2R-expressing tumor cells and some antitumor activity in heavily pretreated patients. However, development was discontinued due to limited efficacy compared to newer agents and immunogenicity concerns[1][3][4][5][6].
