Overview

This is a combination of three drugs: **dabrafenib**, a selective BRAF kinase inhibitor; **trametinib**, a selective MEK1/MEK2 inhibitor; and **clofarabine**, a purine nucleoside antimetabolite. Dabrafenib and trametinib target the MAPK/ERK signaling pathway, primarily used in cancers harboring BRAF V600 mutations. This dual inhibition helps overcome resistance seen with single-agent therapy, resulting in improved outcomes in melanoma, NSCLC, and other BRAF V600E mutation-positive cancers. Clofarabine inhibits DNA synthesis and repair by interfering with ribonucleotide reductase and DNA polymerases, and is principally used in pediatric acute lymphoblastic leukemia but is studied in other malignancies. The rationale for triple therapy may involve combining targeted inhibition of MAPK pathway (dabrafenib + trametinib) with the cytotoxic effect of clofarabine to improve efficacy or overcome resistance, potentially in relapsed or refractory hematologic or solid tumor malignancies.
