Overview

DAC-1522 is a novel Trop2-targeting degrader-antibody conjugate (DAC) designed for the treatment of TROP2-positive malignancies. It consists of a monoclonal antibody targeting Trophoblast cell-surface antigen 2 (TROP2) conjugated to a BET (Bromodomain and Extra-Terminal domain) protein degrader payload. By integrating the selective targeting of an antibody with the mechanistic versatility of protein degradation, DAC-1522 aims to overcome resistance associated with conventional topoisomerase I inhibitor-based antibody-drug conjugates (ADCs) like Enhertu and SKB264. Preclinical studies have demonstrated potent, TROP2-dependent cytotoxicity across various cancer cell lines and robust antitumor activity in xenograft models, including those resistant to standard-of-care ADCs.
